A mathematical model for Herpes Simplex Virus type 1 (HSV-1) reproduction

Herpes viridae, which includes important human pathogens such as Herpes simplex virus type 1(HSV-1), type 2, cytomegalovirus, Epstein-Barr virus, Human herpesvirus-6, 7 and 8, is the major group of DNA virus. All genomic sequence of HSV-1 is reported in 1998. The genome of HSV-1 is approximately 152kbp in size. Total of 74 genes encoding distinct proteins have been identified. The genes of HSV-1 are clssified into three groups, immediate early (IE), early (E) and late genes (L) by the regulation of their expression. Immediate early genes are transcribed immdeiately after infection without new synthesis of other viral proteins. VP-16 within a tegument of infecting viral particle are delivered to the host cell. VP-16 form the transcriptional complex with host proteins, Oct-1 and HCF-1, on the immediate early promoter. This transcriptional complex activates the expression of immediate early gene. Immediate early gene products regulate the expression of the early and late genes directly or indirectly. Early genes are involved in the DNA replication and nucleic acid metabolism. The remaining genes are classified as late genes, and most of these are virion components such as capsid proteins, tegument proteins and envelop proteins. Transcription of IE genes results in the cascade of gene transcription, in which early genes and late genes are transcribed in an ordered temporal pattern. It is known that this genetic cascade plays a critical role in regulating the HSV-1 replication. When HSV-1 infects human cells, it is able to enter two modes of infection , lytic and latent. The lytic mode of HSV-1 infection naturally occurs in epithelial cells. HSV-1 is acquired early in life from oral mucosa. Upon primary infection, HSV first grows lytically in the epithelial cells. Replication of HSV in the epithelial cells results in virus gaining access to sensory nerve terminals, followed by retrograde axonal transport to neuronal cell bodies where latency is established. The latent mode naturally occurs in sensory neuron. Little is known of how the choice between lytic and latent infection is made. We construct a mathematical model for HSV-1 reproduction. The feature of this model is that it is able to analyze the dynamics of viral molecules in one cell. In this seminar, we will show the model and results and disscuss the biological implications.

ヘルペスウイルス属は哺乳類に広く感染するDNAウイルスで、HSV-1, HSV-2, Epstein-Barr virus, Cytomegalo virus, Human herpes virus-6, 7, 8など人の病原ウイルスとしてもよく知られたウイルスが属する。HSV-1は152kbpのdouble stranded DNAを持つDNAウイルスである。HSV-1の全ゲノム配列は1998年に報告されており、74個の蛋白質がコードされていることが知られている。 HSVー1遺伝子は発現時期によってImmediate early gene (IE), Early gene (E), Late gene (L)の3つのグループに分類される。細胞内にウイルスが侵入すると30分以内にIE geneの発現が開始される。ウイルス粒子内に含まれるVP-16蛋白質はホスト細胞のタンパク質Oct-1, HCF-1と転写複合体を形成する。この転写複合体によりIE gene promoterが活性化されるので、IE geneの発現には新規のタンパク合成は必要とされない。IE gene遺伝子産物はウイルス遺伝子の転写を直接、間接に活性化し、early geneの発現が、続いてlate geneの発現が開始される。E geneの遺伝子産物は主にDNA polymerase subunitと核酸代謝に関わる酵素で、ウイルス遺伝子の複製に関与している。一方、L gene遺伝子産物はenvelopやcapsidなどウイルス粒子の外郭を構成する。このウイルス遺伝子カスケード(IE→E→L)はウイルス粒子複 製を調節するのに重要な役割を果たしていることが知られている。HSV-1は初回感染では口腔粘膜から感染し、細胞内で増殖する。その後細胞を破壊して 周囲へと拡がって行く。これが通常のlytic サイクルである。一方、軸索から逆行的に神経細胞に到達したHSV-1は潜伏期へ入る。潜伏期ではウイルス粒子の複製は停止してしまう。lyticサイ クルと潜伏期を切替えるメカニズムはまだよく解かっていない。 今回我々は、細胞内でのウイルス粒子の複製反応をモデル化したので報告する。既存の感染モデルと違い、一細胞内でのウイルス粒子の濃度変化を解析できる のがこのモデルの特徴である。本セミナーでは系の振舞や、各パラメータの依存性、パラメータの持つ生物学的意義などについてディスカッションする予定で ある。
